Childhood obesity may affect timing of puberty, create problems with reproduction

ScienceDaily (July 31, 2012) ? A dramatic increase in childhood obesity in recent decades may have impacts that go beyond the usual health concerns -- it could be disrupting the timing of puberty and ultimately lead to a diminished ability to reproduce, especially in females.

A body of research suggests that obesity could be related to growing problems with infertility, scientists said in a recent review, in addition to a host of other physical and psycho-social concerns. The analysis was published in Frontiers in Endocrinology.

Human bodies may be scrambling to adjust to a problem that is fairly new. For thousands of years of evolution, poor nutrition or starvation were a greater concern, rather than an overabundance of food.

"The issue of so many humans being obese is very recent in evolutionary terms, and since nutritional status is important to reproduction, metabolic syndromes caused by obesity may profoundly affect reproductive capacity," said Patrick Chappell, an assistant professor of veterinary medicine at Oregon State University and an author of the recent report.

"Either extreme of the spectrum, anorexia or obesity, can be associated with reproduction problems," he said.

Researchers are still learning more about the overall impact of obesity on the beginning of puberty and effects on the liver, pancreas and other endocrine glands, Chappell said. While humans show natural variations in pubertal progression, the signals that control this timing are unclear.

But in general, puberty appears to be starting earlier in girls. It is being accelerated.

This may have several effects, scientists have found. One theory is an impact on kisspeptin, a recently characterized neurohormone necessary for reproduction. Normal secretions of this hormone may be disrupted by endocrine signals from fat that serve to communicate to the brain.

Another possible affect on pubertal timing, and reproduction in general, is disruption of circadian clocks, which reflect the natural rhythms of night and day. Disrupted sleep-wake cycles can affect the secretion of hormones such as cortisol, testosterone, and insulin, researchers have found.

"Any disruption of circadian clocks throughout the body can cause a number of problems, and major changes in diet and metabolism can affect these cellular clocks," Chappell said. "Disruption of the clock through diet can even feed into a further disruption of normal metabolism, making the damage worse, as well as affecting sleep and reproduction."

Molecular mechanisms have only started to be uncovered in the past decade, the report said, and the triggers that control pubertal development are still widely debated. For millennia, many mammals made adjustments to reduce fertility during periods of famine. But it now appears that an excess of fat can also be contributing to infertility rates and reproductive diseases.

Some studies in humans have found correlations between early puberty and the risk of reproductive cancers, adult-onset diabetes, and metabolic syndrome. Early onset puberty has also been associated with increased rates of depression and anxiety in girls, studies have found, as well as increased delinquent behavior, smoking and early sexual experiences in both girls and boys.

Other research has suggested that such problems can persist into adulthood, along with lower quality of life, higher rates of eating disorders, lower academic achievement and higher rates of substance abuse.

Additional research is needed to better understand the effect of these processes on metabolism, hormones and other development processes, the survey concluded.

Zombies show up to counter Westboro Baptist Church protestors

Hundreds of protestors dressed as zombies turned out to support the troops (KIRO-TV.com)After reading this story, you may begin to see zombies in a whole new light. While they get a lot of criticism over their love of human flesh, it turns out that some zombies also go out of their way to support the troops.

The Westboro Baptist Church had recently announced that it would be holding one of its protests outside the Joint Base Lewis-McChord in DuPont, Washington. But last Friday, the controversial group was met with a far larger group of counter-protestors ? dressed as zombies.

The controversial, Kansas-based church did not explain exactly why it was protesting in DuPont, but they have held a number of similar protests outside of military funerals in objection to gay rights.

That's when Melissa Neace and three of her friends organized the Facebook event "Zombie'ing Westboro Baptist Church AWAY from Fort Lewis!"

"I have decided that the most violent free way to protest back is simple... a zombie apocalypse. So I ask EVERYONE who is able to attend that day, please come dressed in your finest zombie outfit," Neace wrote on the page.

The event page received such a positive response that the zombies outnumbered the protestors by a reported level of 300 to 8, the News Tribune reported.

"We wanted to turn something negative around, into something people could laugh at and poke fun at," Neace told the paper. "It was the easiest way to divert attention from something so hateful."

"I think that their message is very hateful, and Jesus was not a hateful person. He loved everybody," counter-protestor Ashley Winslow told KIRO-TV.

Pitbull gets bear spray during Alaska visit

KODIAK, Alaska (AP) ? Miami rapper Pitbull can now say he's safe from bears in Alaska.

Pitbull received a care package Monday from Walmart during his visit to Kodiak, complete with bear repellent spray and bear bells used by hikers to make noise, the Kodiak Daily Mirror reported.

The bear spray had special meaning to about 250 people who attended the Pitbull appearance at the Coast Guard base in Kodiak. The rapper had tweeted he heard the local Walmart sold the spray.

In a marketing deal, Pitbull ? whose real name is Armando Christian Perez ? agreed to visit the Walmart that got the most likes on its Facebook page.

Kodiak was the easy winner, thanks to David Thorpe, a Boston writer who thought it would be funny to send Pitbull to the most remote Walmart possible. Thorpe also was in Kodiak Monday and met the rapper.

Pitbull was a sport, receiving a key to the city from Kodiak Mayor Pat Branson, who donned shades to mimic the dapper rapper's look.

Pitbull tweeted a picture of him holding a child onstage alongside Alutiiq (UH'-loo-tic) dancers and said, "Thank U Kodiak ... I am honored, truly."

The rapper, known as "Mr. Worldwide," told the audience he would put Kodiak into a future song.

Following the appearance at the Coast Guard base, he met employees of the Walmart.

8.7 million mobile customers hacked in S.Korea

South Korean police have arrested two hackers who stole personal data of 8.7 million customers of the nation's second-biggest mobile operator, the company said.

KT said the hackers -- formally arrested on Sunday -- had stolen data such as customers' names, phone numbers and residential registration numbers for five months since February and sold the information to telemarketing firms.

"The number of affected people account for nearly a half of about 17 million customers of ours," a KT spokesman told AFP, adding the company had alerted police on July 13 after detecting traces of hacking attacks.

Yonhap news agency, citing police, said the duo -- including a former veteran programmer at a local IT company -- had earned at least 1 billion won (about $880,000) by selling the stolen data.

Seven other people were also booked for buying the leaked data for telemarketing purposes, Yonhap said.

"We deeply bow our head in apology for having your precious personal information leaked... we'll try our best to make such things never happen again," KT said in a statement to customers.

Hacking attacks on major companies aimed to gain access to the personal data of their customers is a frequent occurence in South Korea, one of the world's most-wired nations.

Seoul authorities said in July last year hackers using an Internet address registered in China had gained access to South Korean major websites including web portal Nate.com and may have stolen the private data of 35 million users.

In November 2011, Seoul's top games developer Nexon saw personal information of 13 million users of its popular online game MapleStory stolen by hackers.

In March 2010, authorities launched a probe into the security systems of major retailer Shinsegae and 24 other companies after private data on 20 million customers was leaked.

Garrigus sets record and takes lead in Canada

ANCASTER, Ontario (AP) ? Robert Garrigus would have made Arnold Palmer proud Saturday at the Canadian Open ? for the way he treated the people, and for the scores on his card.

Garrigus combined his power with a couple of timely putts at rain-softened Hamilton for a 6-under 64, breaking the 54-hole scoring record at golf's third-oldest championship that was first set more than a half-century ago by Palmer. Equally impressive was when he didn't even have a club in his hand.

After a routine par on the sixth hole, Garrigus walked over to a volunteer and said, "Thanks for being here." After he chunked a wedge into a bunker on the seventh, one of his poorer shots of the third round, he found another volunteer behind the green and said, "Appreciate what you do for us."

It's the kind of stuff that made Palmer the King.

"It makes me feel good just to say it," Garrigus said. "Like I said, it's on their dime. And we need the fans. We need the volunteers. We need the sponsors, and a lot of guys out there don't lean that way to thank the volunteers. I've had hundreds of volunteers come up to me and say, 'Thanks for saying thank you.' That means a lot."

Now if he can just finish like the King.

Garrigus had a one-shot lead over William McGirt, who played with poise with his name atop the leaderboard for the first time on the weekend at a PGA Tour event. McGirt had a 66 and actually lost ground, going from a share of the lead to one-shot behind. He will be in the last group with Garrigus.

Garrigus was at 16-under 194, one shot better than Palmer in 1955 at Weston Golf & Country Club in Toronto, and matched two years ago by Dean Wilson at St. George's Golf & Country Club, also in Toronto.

"Oops. Sorry, Arnie," Garrigus said when told about the record.

Palmer, however, went on to win the 1955 Canadian Open for the first of his 62 titles on the PGA Tour. Garrigus will be going for his second win, though this tournament is a long way from being settled.

Garrigus was fortunate to escape with par, not to mention his health, on the final hole when his 3-iron off the tee ran out 290 yards and just short of the bridge. Because he was inside the hazard, he couldn't ground the club and had a rules official make sure the bridge wasn't beneath the ball. It was a few inches ahead of the ball.

"If I hit it a millimeter fat, I break my wrist," Garrigus said. "I had to knife it out of that lie and catch part of the bridge, and hopefully the ball gets the top part of the bridge and bounces up. And that's exactly what I did. I hit it perfect."

Scott Piercy had a 67 and was two shots behind.

Scott Stallings, who won last week in Mississippi, birdied his last two holes for a 63 and was four shots behind, along with Chris Kirk (63) and Bo Van Pelt (67). Stallings ended a streak of nine consecutive PGA Tour events in which the 54-hole leader failed to win. The ninth was Ernie Els coming from six shots behind at the British Open. Stalling won later that day.

"A lot of guys haven't been able to hold leads this year," said Garrigus, still known for losing a three-shot lead on the final hole in Memphis two years ago.

No matter the difficulty of any course ? Hamilton rates among the best in the Canadian Open rotation ? soft greens are no match for the best players. And now, the 72-hole record at the Canadian Open is in jeopardy. That belongs to another Palmer ? Johnny Palmer ? who had a 263 in 1952 at St. Charles in Winnipeg, Manitoba.

Garrigus' goal?

"Foot to the floor, as much as possible," he said.

He wasn't speaking about the scoring record as much as leaving town with the trophy, which would put him in a World Golf Championship next week at Firestone and the Masters next April. He might not have a choice the way scoring has been this week.

McGirt, who started the day atop the leaderboard for the first time in his career, was never more than two shots behind. That came on the par-3 13th, when his tee shot came up well short and into a bunker, leading to his lone bogey. He atoned for that with a birdie on the next par 3 at No. 16, and recovered from a poor tee shot with an up-and-down from some 45 yards short of the 18th green.

Garrigus wasted no time joining the chase with a 316-yard tee shot that left him only a 5-iron that he stuffed inside 3 feet on the 530-yard fourth hole. He followed by driving through the green on the 329-yard fourth hole into a back bunker for an up-and-down, then made another strong move around the turn with three birdies in a four-hole stretch, courtesy of a couple of tricky putts.

The third round started late and in threesomes to cope with overnight rain, though Hamilton began to dry out in the afternoon. Proof of that was the 3-iron Garrigus hit off the 18th tee to stay well short of the creek. He was stunned to see it go through the fairway near the bridge.

The forecast Sunday is for sunshine, which could make Hamilton the test it typically is.

Kirk, who began the third round six shots behind. Starting with the third hole, Kirk made seven birdies in an eight-hole stretch. He was 7 under for his round with eight holes remaining, needing birdie on half of them for a 59. He was never thinking about golf's magic number, though, which was a good thing. He closed with eight pars, and he was OK with that.

"I was just trying to keep my head down and keep going," he said. "When you have those days where everything is going in the hole, you make as many as you can."

DIVOTS: Vijay Singh, winless on the PGA Tour since 2008, started the third round three shots behind and had a 69. He was seven shots out of the lead. ... William McGirt and his wife, Sarah, are expecting their first child in January. That might make it tough for him to get to Kapalua if he were to win on Sunday, a nice problem to have. ... Greenbrier winner Ted Potter Jr. was in the mix until a 40 on the back nine.

While not an official, legal symbol, the copyright symbol with a line through it has become the generally accepted way to indicate that a work is in the public domain. It is usually accompanied by one of the following two statements:

1. This image has been released into the public domain by the copyright holder, its copyright has expired, or it is ineligible for copyright. This applies worldwide.

2. I, the creator of this work, hereby release it into the public domain. This applies worldwide. In case this is not legally possible, I grant anyone the right to use this work for any purpose, without any conditions, unless such conditions are required by law.

When copyright or other intellectual property restrictions expire, works will enter the public domain and may be used by anyone.
More specifically, the public domain comprises the body of knowledge and innovation, especially creative works such as writing, art, music, and inventions, in relation to which no person or other legal entity can

However, intellectual property rights may exist in translations or new formulations of these works.

In addition, works of the United States government are excluded from copyright law and may therefore be considered to be in the public domain. For example, works created by a U.S. government agency become part of the public domain at the moment of creation. Other examples include: NASA photographs, military journalism, federal court opinions (but not necessarily state court opinions), congressional committee reports, census data, and the U.S. government website. Availability of such documents may, however, be limited by secrecy laws.

In the United States, all copyrights and patents have a finite term; when this term expires, the work or invention is released into the public domain. For example, patents expire 20 years after they are filed. However, a trademark registration may be renewed and remain in force indefinitely provided the trademark is used, but could otherwise become generic. Copyrights are more complex; generally, they expire in all countries when a variety of specific conditions are satisfied.

Before 1978, unpublished works were not covered by the federal copyright act. This does not mean that the works were in the public domain; it means that they were covered under state copyright acts. Websites that claim that ?pre-1923 works are safe? are wrong. These works, now under federal copyright, will not expire for several more decades.

Patents

Publishing the details of an invention before applying for a patent will generally place an invention in the public domain and prevent its subsequent patenting by others. For example, when a chemistry journal publishes a formula, this prevents patenting the formula by anyone. There is an exception to this; however, in U.S. law, the rightful inventor may file a patent claim up to one year after publishing it.

Trade Secrets

If guarded properly, trade secrets never enter the public domain. For example, a business may keep a product formula like that for Coca-Cola a secret indefinitely. However, if it is disclosed to the public, the former secret enters the public domain, although the former secret may still be patented in the United States.

Trademarks

A trademark registration is renewable. A trademark owner may maintain a registration indefinitely by paying renewal fees, using the trademark and defending the registration.

However, a trademark or brand may become a generic term for a particular type of product or service if people do not use it as a trademark (i.e. as a name or graphic representation exclusively identifying that that product or service originates from a particular business).

A genericized trademark ? sometimes known as a generic trade mark, generic descriptor or proprietary eponym ? is a trademark or brand name which has become synonymous with the general or formal term for a particular type of product or service, to the extent that it often replaces this term in colloquial usage.

One famous U.S, example is ?thermos?. An interesting philosophical issue is that a manufacturer who invents an amazing breakthrough product which cannot be succinctly described in plain English (for example, a vacuum-insulated drinking flask) will inevitably find its product described by the trademark (?Thermos?). If the product continues to dominate the market, eventually the trademark will become generic (?thermos?). Other examples are: escalator, trampoline, raisin bran, linoleum, dry ice, shredded wheat, mimeograph, yo-yo, nylon, kerosene, cornflakes, cube steak, lanolin, and high octane, masonite, allen wrench, pabulum, styrofoam, and zipper.

One consequence of a trademark becoming generic is that the exclusive rights which may attach to the use or registration of the trademark can no longer be legally enforced. Genericide typically occurs over a period of time where the trademark owner does not maintain or enforce its proprietary rights (e.g. by using the mark or by pursuing infringement action).
However, genericide is not an inevitable process. In the late 1980s ?Nintendo? was becoming synonymous with home video game consoles but Nintendo was able to reverse this process through marketing campaigns. Xerox was also successful in avoiding its name becoming synonymous with the act of photocopying.

Domain Names

A domain name, which may be bought and sold, never enters public domain. If nobody owns it, it simply doesn?t exist. Top level domains, such as .com, are controlled by the Internet Corporation for Assigned Names and Numbers.

Many people are using the Internet to contribute to the public domain, or make works in the public domain more accessible to more people. For example, Project Gutenberg is coordinating the efforts of people who transcribe works in the public domain into electronic form in order to create e-book. New research method provides better insights into the world of microbes

ScienceDaily (July 27, 2012) ? A team of Luxembourg-based researchers, working at the Luxembourg Centre for Systems Biomedicine, has developed a research method that will allow scientists to study microbes in more depth than ever before.

Microbes, such as bacteria, fungi and viruses, are invisible to the human eye but have been identified just about everywhere on earth: in soil, water, and air. Many different types of microbes usually live closely together in 'microbial communities'.

Microbial communities also reside in the human body, which is home to trillions of these tiny organisms. Biologists have long suggested that microbes in the human body can cause or contribute to some human diseases, such as diabetes. The new research method might soon be able to prove this hypothesis.

"In order for us to understand the impact that microbes might have on human health, we need to be able to measure the biomolecular information contained within the DNA, RNA (ribonucleic acid), proteins, and small molecules of microbes in a truly systematic way. This was not possible until now," explains Dr. Paul Wilmes, who runs the Eco-Systems Biology lab at the LCSB.

Using the method described by Wilmes, researchers can, for the first time, measure and integrate all the important biomolecular information from a single sample, providing scientists with high-resolution molecular snapshots of microbial communities in, for example, the human stomach and intestine.

The research method developed by Wilmes can also be applied to microbial communities found in other environments. In this particular study, authors used, for example, microbes that grow in wastewater treatment plants and that accumulate large amounts of high-energy fats. By using Wilmes' new research method, scientist can do in-depth studies on these communities. "Once these microbial communities are better understood, we might be able to exploit these communities for the comprehensive reclamation of energy-rich fats from wastewater," says Dr. Paul Wilmes.

This work was carried out within the realm of a Luxembourg National Research Fund ATTRACT grant.

Russia warns of looming 'tragedy' in Aleppo

Russia warned Saturday that a "tragedy" was looming in Syria's second city of Aleppo but said it was unrealistic to expect Damascus to stand by when armed rebels were occupying major cities.

"We are persuading the government that they need to make some first gestures," Foreign Minister Sergei Lavrov told a news conference alongside his Japanese counterpart.

"But when the armed opposition are occupying cities like Aleppo, where yet another tragedy is brewing, as I understand ... it is not realistic to expect that they (the government) will accept this," Lavrov added.

The Syrian army launched a fightback against rebels in Aleppo on Saturday amid concern among Western governments about reprisals against the civilian population of the country's second city.

"How can you hope that in such a situation, the government will simply reconcile itself and say, 'All right, I was wrong. Come on and topple me, change the regime'?" Lavrov asked rhetorically.

"It's just not realistic -- not because we are holding onto this regime -- but it simply doesn't work," he told the news conference in the southern city of Sochi which was broadcast live by state media.

In Syria, "excesses are committed from all sides. ... We need to put pressure on all of them," Lavrov said, accusing Western countries of providing assistance to the opposition fighters.

"Our Western partners ... together with some of Syria's neighbours are essentially encouraging, supporting and directing an armed struggle against the regime."

"The price of this is yet more blood," he said.

Russia has repeatedly rejected accusations that Moscow is backing the regime of President Bashar al-Assad in the conflict, claiming to have an even-handed approach while rebuking the West for siding with the rebels.

Lavrov also told reporters on Saturday there were meetings planned in the near future with Syrian rebels "in Russia and foreign countries where these boys are based," and that the point driven home would be "that if it is a revolution, they must not seek the backing of the UN Security Council".

When asked later Saturday if Russia would provide Assad with asylum, he said: "We are not even thinking of that," Russian news agencies reported.

Last week Russia along with China for the third time vetoed a UN Security Council resolution on Syria that would have threatened sanctions against Assad, to the outrage of Western nations.

The economic impact of the Olympics has not been the same for all host countries. According to the Olympic Studies Centre at the Autonomous University of Barcelona, the games held in the Catalonian capital 20 years ago were a resounding and incomparable success. But it is not always the case.

The Olympic Games have always been thought of as a great opportunity to give a long-term boost to the economy by taking advantage of a short-term event. But before embarking on the costly effort of organising the Olympics, each candidate city should evaluate their own goals and capacities in order to avoid failure. This is the main conclusion of a report by the Olympic Studies Centre at the Autonomous University of Barcelona (CEO-UAB), in which the economic impact of the Olympics from Tokyo '64 to the present day was studied.

"The overall balance of celebrating the games tends to be positive. There are three obvious cases of failure: that of Montreal, due to financial causes, perhaps because all that investment was a little premature; that of Atlanta, because they tried to organise the games wit no urban impact, and in addition there was a terrorist attack; and Athens, where the organisation failed and the games bore little fruit", said to SINC one of the CEO researchers, Ferran Brunet.

Those celebrated in Barcelona are considered to be an example of profitability. One of the causes of its economic success was that the building work related to sport made up only 9.1% of the total investment connected with the games. "Such a small proportion can be explained by the huge amount of private investment which was triggered by the boost of the Olympics on the city", the CEO research indicates.

61.5% of Olympic investment was targeted to civil engineering, pointing to a key aspect of Barcelona '92: its capacity to structure the city. The construction of the Barcelona circulars various ring-road axes , the reopening of the city to the sea through the construction of the Olympic village, and the creation of various new centrality areas and the Olympic areas of Montjuc, Diagonal and Vall d'Hebron were the main developments.

The city not only reacted very well to the games, but also knew how to maintain the expansion in an unprecedented manner", states the expert.

In fact, the Olympic investment and its economic impact in Barcelona are not comparable with any other host city of the Olympic Games. Only Tokyo 1964 reached even half the investment impulse of Barcelona. In Seoul 1988 there was significant investment, but in Atlanta 1996 and Sydney 2000 it was very limited. Athens 2004 tried it and Beijing 2008 followed the Barcelona 1992 model with respect to deep urban and economic impact, as will Rio 2016.

In the case of London 2012 their objective is to improve their strategic position among the global regions. "It is a city which is already 'made' and with respect to Europe it is the top region for the location of businesses from the 80s to the present. The relative impact of London 2012 will be lower than in Barcelona, but the games will have a large influence on the visibility of the city during the coming years and will have a huge effect, although to a large extent intangible, on improving its competitiveness", highlights the researcher.

How a single brain trauma may lead to Alzheimer's disease

ScienceDaily (July 24, 2012) ? A study, performed in mice and utilizing post-mortem samples of brains from patients with Alzheimer's disease, found that a single event of a moderate-to-severe traumatic brain injury (TBI) can disrupt proteins that regulate an enzyme associated with Alzheimer's. The paper, published in The Journal of Neuroscience, identifies the complex mechanisms that result in a rapid and robust post-injury elevation of the enzyme, BACE1, in the brain. These results may lead to the development of a drug treatment that targets this mechanism to slow the progression of Alzheimer's disease.

"A moderate-to-severe TBI, or head trauma, is one of the strongest environmental risk factors for Alzheimer's disease. A serious TBI can lead to a dysfunction in the regulation of the enzyme BACE1. Elevations of this enzyme cause elevated levels of amyloid-beta, the key component of brain plaques associated with senility and Alzheimer's disease," said first author Kendall Walker, PhD, postdoctoral associate in the department of neuroscience at Tufts University School of Medicine (TUSM).

Building on her previous work, neuroscientist Giuseppina Tesco, MD, PhD, of Tufts University School of Medicine (TUSM), led a research team that first used an in vivo model to determine how a single episode of TBI could alter the brain. In the acute phase (first two days) following injury, levels of two intracellular trafficking proteins (GGA1 and GGA3) were reduced, and an elevation of BACE1 enzyme level was observed.

Next, in an analysis of post-mortem brain samples from patients with Alzheimer's disease, the researchers found that GGA1 and GGA3 levels were reduced while BACE1 levels were elevated in the brains of Alzheimer's disease patients compared to the brains of people without Alzheimer's disease, suggesting a possible inverse association.

In an additional experiment using a mouse strain genetically modified to express the reduced level of GGA3 that was observed in the brains of Alzheimer's disease patients, the team found that one week following traumatic brain injury, BACE1 and amyloid-beta levels remained elevated even when GGA1 levels had returned to normal. The research suggests that reduced levels of GGA3 were solely responsible for the increase in BACE 1 levels and therefore the sustained amyloid-beta production observed in the sub-acute phase, or seven days, after injury.

"When the proteins are at normal levels, they work as a clean-up crew for the brain by regulating the removal of BACE1 enzymes and facilitating their transport to lysosomes within brain cells, an area of the cell that breaks down and removes excess cellular material. BACE1 enzyme levels may be stabilized when levels of the two proteins are low, likely caused by an interruption in the natural disposal process of the enzyme," said Tesco, assistant professor of neuroscience at Tufts School of Medicine and member of the neuroscience program faculty at the Sackler School of Graduate Biomedical Sciences at Tufts.

"We found that GGA1 and GGA3 act synergistically to regulate BACE1 post-injury. The identification of this interaction may provide a drug target to therapeutically regulate the BACE1 enzyme and reduce the deposition of amyloid-beta in Alzheimer's patients," she continued. "Our next steps are to confirm these findings in post-mortem brain samples from patients with moderate-to-severe traumatic brain injuries."

Moderate-to-severe TBIs are caused most often by traumas, such as severe falls or motor vehicle accidents, that result in a loss of consciousness. Not all traumas to the head result in a TBI. According to the Centers for Disease Control and Prevention, each year 1.7 million people sustain a TBI. Concussions, the mildest form of a TBI, account for about 75% of all TBIs. Studies have linked repeated head trauma to brain disease and some previous studies have linked single events of brain trauma to brain disease, such as Alzheimer's. Alzheimer's disease currently affects as many as 5.1 million Americans and is the most common cause of dementia in adults age 65 and over.

Additional authors on the study are Eugene Kang, MPH, research assistant in the department of neuroscience at TUSM; Michael Whalen, MD, PhD, Neuroscience Center and department of pediatrics at Massachusetts General Hospital and associate professor at Harvard Medical School; and Yong Shen, MD, PhD, of the Center for Advanced Therapeutic Strategies for Brain Disorders at Roskamp Institute.

Bird Flu Researchers To Meet About Research Moratorium

Top influenza researchers around the world published a statement back in January saying they would temporarily hold off on any work with contagious, lab-altered forms of a particularly worrisome form of bird flu.

The unusual voluntary moratorium was supposed to last only 60 days, but it's been more than six months. And scientists don't agree on what should happen next.

Some scientists and researchers say these mutant bird flu viruses could cause a devastating pandemic if they ever got out of the lab. Others argue that the work is vital to help public health officials get ready for the possible threat of a flu pandemic that might emerge naturally, as bird flu viruses mutate in the wild.

Flu researchers are going to New York next week for the annual conference of the government-funded Centers of Excellence for Influenza Research and Surveillance (CEIRS). Researchers who made the mutant viruses will be there, plus others who signed the voluntary moratorium.

At a recent press briefing, Anthony Fauci, director of the National Institute of Allergy and Infectious Diseases (NIAID), which funded the controversial experiments, said that he intended to go to the meeting and "discuss in some detail with the group the kinds of approaches we can have to try and expedite as quickly as possible the lifting of the moratorium."

But some ask, what is the hurry? David Relman, a microbiologist at Stanford University who was part of the group that advised the government on whether the details of the bird flu experiments should be made public, believes that "there is no particular reason to rush ahead right now, with what might be a very substantial kind of risk."

Relman says if the moratorium was lifted right after the upcoming meeting, "I'd be concerned."

He's not the only one who feels that way. Marc Lipsitch, an epidemiologist at the Harvard School of Public Health, says that research on these mutant viruses is potentially valuable. "But the risks are not to scientists. The risks are to the world," he says. "The risks are that one of these viruses gets out of a laboratory and starts to spread from person to person. And so the people who have something at stake are not the scientific community only."

He thinks there needs to be a much broader conversation than has happened so far.

"I'm a little disappointed that it hasn't been a very public discussion," says Lipsitch, who feels it's been mostly been within the flu research community.

When asked to respond to this type of concern, Yoshihiro Kawaoka of the University of Wisconsin-Madison, whose lab did one of the recent bird flu experiments, said via email that "I recommend that those who have concerns should voice their opinions to NIH directly, rather than through the media."

When an NPR reporter called NIAID to ask about attending next week's conference to report on the discussion, a spokesperson said the agency was not running the meeting and that it was up to the flu researchers. Adolfo Garcia-Sastre, a virologist at Mount Sinai School of Medicine who is one of the organizers, said "if we open the meeting to reporters, then there will be too many people. I don't think we can do that."

He said he doubted the moratorium would be lifted at the meeting, because it was really not up to the flu community. He says flu researchers are still waiting for official word from the U.S. government about what kind of lab safety will be required for federally funded work, and what kinds of risk-mitigation plans will have to be submitted before experiments get done. All of that needs to happen before flu researchers could go ahead.

Meanwhile, NIAID's Fauci says that when he talked about trying to "expedite as quickly as possible the lifting of the moratorium," he didn't mean to lift it quickly.

"I really meant the expediting of a frank, open, clear discussion, instead of, you know, press-type sound bites back and forth. That's what I meant by expedite. Not that I wanted the moratorium lifted the day after the CEIRS meeting," Fauci told NPR. "That's not what I meant."

Asked how the discussion could be open if the meeting was closed, Fauci replied that "you have to first start off with one step at a time. If the scientific community isn't even in agreement or even able to articulate with each other what it is they want to do, how are they going to express it in a way that the public can understand what it is they want to do?"

But this kind of approach doesn't sit well with folks who want more transparency.

"There are plenty of people in the public who are not stupid and are perfectly well able to understand the issues that are involved here and to decide whether or not they think this is a good use of taxpayers' money," says Richard Roberts, a Nobel Prize-winning molecular biologist at a company called New England Biolabs.

He thinks the moratorium should be continued "until there has been a comprehensive debate about what should happen, not just some behind-closed-doors debate by a group of insiders who have a vested interest in seeing this research continue."

By that Roberts means not just the flu researchers, but the government agencies that funded their work. He says officials need the outside world to think they made good decisions. "They have such an obvious conflict of interest that they're not able to represent the public," says Roberts.

Fauci strongly disagrees, saying the National Institutes of Health has no conflict of interest here.

"I mean that's a nice catchword, 'conflict of interest.' But the main mandate is that we really don't have any vested interest. We want to do the right thing," says Fauci. "These experiments were done for the thought of protecting the global health."

At the time that the experiments were done, says Fauci, it was felt by program officials and by everybody else involved that the benefits of doing ? and openly publishing ? the experiments outweighed the risks. And he points out that a panel of advisers to the government ultimately agreed with that view, saying the work should be published in full.

The public will be able to weigh in on these issues soon, says Fauci, when officials solicit comments on a new plan for overseeing certain kinds of experiments with high-risk pathogens.

But some flu experts say there are bigger questions here that go beyond what's done in any one country ? and that they need to be addressed at an international level.

"The way that I see it is that all the discussions that are ongoing are not really hitting the nail on the head," says Ilaria Capua, an Italian flu researcher at the Istituto Zooprofilattico Sperimentale delle Venezie, Legnaro, who signed the moratorium.

In her view, if this kind of work continues to get funded and published in the U.S. and Europe, the technology for making mutant flu viruses will get easier and easier, and more and more labs will want to do it.

"And then we have 100, 200 labs around the world which contain in their freezers influenza viruses of high pathogenicity which are transmissible in humans," says Capua. "Is this what we want?"

She notes that this proliferation could increase the risk of a lab accident, especially if work starts getting done in less stable regions of the world where political turmoil or natural disasters could compromise security.

The World Health Organization is planning to have an open meeting next year that will address some of these issues. In the meantime, that agency has just released some guidelines for appropriate risk-control measures for research with these mutant bird flu viruses.

"Given the potential of these newly developed laboratory-modified H5N1 strains to start a pandemic," the guidance notes, "it is important that facilities that are NOT able to identify and appropriately control the risks associated with these agents REFRAIN from working with them."
